His conversation is with his dead puppy. He asks the puppy why it died; Lennie, because he has never really understood his own strength, believes that he did not bounce the puppy too hard. He also assumes that because the puppy is larger than mice, it should not die as easily as a mouse might. It reflects his thinking about other things that he has killed with his strength.
