We will expect to see the isolated colonies in area 4. This is due to the fact that streak plating method is used, which isolates the strains in a step by step method and gives the best results. The inoculating loop is first dip in the sample and then streaks are made on first plate. This process is repeated three more times, but every time dipping the loop in next plate, which ensures best isolation.
