1. Endospores are formed by bacteria to enable survival in hostile conditions. 2. The tough protein coat - comprised of keratin - which an endospore is made of makes it very resistant to normal staining procedures. 3. It is necessary to infuse the cells with malachite green, using heat to disrupt the endospores while doing so. 4. Malachite green is water-soluble, and will not adhere to the cell, so it will easily stain the disrupted endospores.
