In a protein crystal, the first few residues of the polypeptide chain may experience fewer intermolecular contacts or be innately mobile. These residues are more motile and less ordered i.e they do not occupy a set position in the crystal. Because of the disorder, they fails to generate a coherent diffraction pattern when hit by the x-ray beams and without a diffraction pattern the electron density cannot be generated.
