Asp 101 and Arg 114 form Hydrogen bonds with the substrate molecules.Ala cannot form these hydrogen bonds,so the substituted enzyme is less active. The effect on lysozyme's activity of mutating Glu 35 to Asp and Asp 32 to Glu would diminish lysozyme's activity because removal or addition of a removal of a methylene group would move the Glu and Asp carboxyl group from their catalytically most effective positions. Lysozyme is responsible for hydrolysis of glycosidic bond between NAM and NAG in bacterial cell wall.Lyzozyme contains Glu 35 and Asp 52 in active site. Glu 35 cleaves C-0 bond in substrate by transferring proton to polar O atom of glycosidic bond. Asp 52 stabilises oxonium ion.
